    I don't use Ai a lot, but the shape-builder tool is pretty cool !

    The transform in effects menu.
    Just take your A5 artwork/design, put it in a SRA3 sheet. Select it, go to effects>transform and make a copy moving it 153mm to the right. Take again the original artwork/design and apply another transform to it, this time make a copy moving it 215 mm down.
    You have 4 copies of the design in the sheet. But, the amazing thing is that you can change font, text, color to the original and all the copies will update at the same time.
    It's kinda "speedy gonzales" on digital printing!!!

    There are so many cool features in Illustrator that it's hard to think of just one. I particularly like the stroke width tool, that I totally loved from its introduction back in CS5. It's just so easy to use and you have lots of control if you need a variable width stroke. In CS4 and before, you had to make two strokes, and modify them both when you needed to change the curviness, length etc.

    effects like drop shadow that OP stated are actually raster, not vector so thats kinda lame if you are a vector freak. there are other ways of achieving this effect in vector.
    my fav tools are...pen tool, pathfinder,paintbrush tool

    I realize it's not a native AI tool but...

    The Vectorscribe and Inkscribe plug-ins are by far the tools I use the most. To all those who cited the pen tool as their favorite, your mind will be blown by these two plugins. You're welcome.
